line art layer selection

line art layer selection
I'd like to draw a line art in a layer and than create a second layer for coloring it, so i don't have to go around cleaning up white dots near my lines inbetween for exapmle my brown color. So now is there a way to select the space inbetween shapes i make in one layer and color them in, in a second one?
It's easy enough to just manually color around the edges, in the second layer, when the lines are thick enough so i don't have to nervously spend time making sure colors aren't overlapping, but this is quite impractical when doing 1 pixel broad lines.
