I need a way to quantify the color intensity of redness in a picture.

I did an experiment where I stained the lignin in the roots of plants, after treating them with one of three different treatments, and then took pictures of the redness with a microscope. I need a way to quantitatively show that the level of ligninification is different in response to different treatments.

I have tried a few different approaches in ImageJ, like using the thresholding tool to determine the area that is occupied. This unfortunately didn't end up working well, and its not necessarily the most direct method. I also tried separating the channels from each other, but that also did not help.

I started trying to use GIMP, by using the average %RGB occupied in a 20*20 selection with the average color picker. I'm new to the software and I don't know much about this sort of thing in general, so any help or pointers are appreciated, for GIMP or any other software. There are about 600 total pictures that I need to determine a quantity of red for, so preferably the method wouldn't require too many steps, etc.

I attached two sample pictures of what I'm working with. Obviously one picture has very low lignification, and the other picture has much more lignification, and in turn is much more red.

What about making it readonly, so that people won't register only to find spam and spammers won't flood the useful informations ?
